The role of tumour necrosis factor-a and interleukin-1 as mediators of tissue injury during sepsis from faecal peritonitis and immunomodulation with pentoxifylline.
Abstract
THE LOCATION OF TUMOUR NECROSIS FACTOR ALPHA AND INTERLEUKIN-1 PRODUCTION DURING SEPSIS DUE TO PERITONITIS IS NOT KNOWN.A MULTITUTE OF POSSIBLE ADVERSE ACTIONS OF THESE TWO PROINFLAMMATORY CYTOKINES HAS BEEN SHOWN IN ANIMAL MODELS OF ACUTE INTRAVASCULAR SEPSIS, BUT THESE HAVE NOT BEEN SUBSTANTIATED IN CLINICALLY RELEVANT MODELS OF PERITONITIS OR HUMAN TRIALS.IN THIS STUDY, USING THE CAECAL LIGATION AND PUNCTURE MODEL IN THE MOUSE AND DIFFERENTIAL RT-PCR TO COMPARE TNF MRNA OR IL-1 MRNA TISSUE LEVELS IN CONTROL AND SEPTIC MICE, WE FIRST SHOW THAT THERE IS A COMPARTMENTALISATION OF TNF AND IL-1B PRODUCTION DURING FAECAL PERITONITIS.WHILE CYTOKINE SERUM LEVELS REMAIN RELATIVELY LOW FOR TNF AND UNDETECTABLE FOR IL-1, LUNG TISSUE RESPONDED MORE VIGOROUSLY THAN LIVER,SPLEEN AND INTESTINE IN TERMS ΟF TNF MRNA AND IL-1B MRNA EXPRESSION.MAJOR MECHANISMS BY WHICH TNF AND IL-1 HAVE BEEN REPORTED TO INDUCE TISSUE DAMAGE, INLCUDE PROMOTION OF NEUTROPHIL SEQUESTRATION AND REDUCED MICROVASCULAR ...
