T-cell apoptosis in the CNS is an effective mechanism for the noninflammatory, non-phlogistic resolution of autoimmune T-cell infiltrates. Ingestion of apoptotic leukocytes by microglia results in an efficient clearance of the inflammatory infiltrate, followed by a downregulation of proinflammatory phagocyte immune functions. We investigated the effects of different immunomodulatory agents on Lewis rat microglial phagocytosis of apoptotic autologous thymocytes and of myelin-basic protein (MBP) specific, encephalitogenic T-cells. Pretreatment of microglia was obtained by agents that are used for therapeutical purposes in CNS autoimmune diseases like multiple sclerosis. The immunomodulatory agents are 7S immunoglobulin (IVIg), F(ab)₂-fragments of IVIg, glatiramer acetate, methylprednisolone and interferon-beta. The in vitro dosages corresponded to the in vivo used dosages. Pretreatment of microglia with polyclonal 7S immunoglobulins decreased the phagocytosis of apoptotic thymocytes and ...
